測試方法及其原理

國家藥典委員會發布的《中華人民共和國藥典2010 版》二部中推薦采用在線和

離線兩種測試方法,還提供了系統適應性試驗的操作方法。同時對測試總有機碳的儀

器也提出了要求,即首先要能區分無機碳和有機碳;并能排除無機碳對總有機碳的影

響;其次應滿足系統適應性試驗的要求;最后儀器應具有足夠的檢測靈敏度。

要檢測樣品中的有機物濃度,必須將有機物分子分解并且轉化成能夠測量的單

分子形式,這樣就必須把有機物氧化成二氧化碳,并對生成的二氧化碳進行測量。目

前氧化的方法有四種:一、燃燒法;二、光氧化法;三、濕法氧化;四、光化學法。

氧化后有機碳測試的方法有差減法和直接法兩種。

其中差減法的表達式為TOC=TC-IC,即總有機碳是總碳(TC)與無機碳(IC)的差值。

TA-1.0 分析儀采用的氧化方法是光氧化法:

H2O hn(185nm)‐>OH? H? OH? 有機物‐>CO2 H2O

二氧化碳的測量方式是差減法:TOC = TC- IC

系統適應性驗證

系統適應性驗證是一種檢驗TOC 分析儀氧化能力的試驗方法,其計算公式如下:

rss —— 為 1-4 對苯醌對照品溶液的響應值;

rs —— 為蔗糖對照品溶液的響應值;

 分流器

水樣進入儀器后分成相同流量的兩路,其中一路通過延遲線圈進入二氧化碳傳感器,檢測TIC,另一路通過氧化反應器利用紫外燈(UV燈)加二氧化鈦薄膜光催化氧化作用將有機物分解為二氧化碳,進入二氧化碳傳感器檢測TC??傆袡C碳可通過這個差值計算得到:TOC = TC–TIC。

氧化反應器

儀器利用UV射線在二氧化鈦光催化劑的作用下將有機化合物氧化成二氧化碳,氧化反應器是一個UV燈外包螺旋形的石英管。UV燈發出185nm和254nm的光線,使水產生光分解。

UV燈的使用壽命為6個月,當更換時間到期時儀器將出現警告信息,提醒用戶更換UV燈。
